Predict your protein's ferritin encapsulation regime

Upload a PDB structure (or fetch one by ID) to estimate whether a cargo protein is likely to undergo efficient luminal encapsulation (Regime I), steric/structural accommodation (Regime II), or charge-driven off-pathway assembly (Regime III) in apoferritin nanocages, based on net charge, size, and cavity occupancy descriptors.

Advanced options (optional)

Volume is estimated with a fast grid-based approximation by default. For manuscript-consistent numbers, supply a ChimeraX solvent-excluded volume and/or a Prot pi net charge above.

enCAGE is a heuristic design aid, not a validated classifier. Regime boundaries are calibrated on a small in-house cargo panel (equine apoferritin, pH cycling) and should be refined as more data accrue.
